In the 1980s, scientists developed a technique for measuring and comparing DNA called restriction fragment length polymorphism, or RFLP. With RFLP, the laboratory looks at specific sections of DNA extracted from blood samples. By comparing the child with the mother and father, RFLP can eliminate, or exclude, a possible father or include him as the biological father of the child.

However, RFLP requires a large sample of blood and takes a long time to process. DNA testing laboratories today prefer a quicker, simpler method of paternity testing.
